X-Def is better for items IMO while LQ is way better for XP. X-Defense is amusing for awhile but it can get tedious just like LQs. A healthy mix of everything has been working for me.

The leveling gets easier after you have some characters leveled already. I have around 60% bonus xp from having IM to lvl 60, Cyclops lvl 50 synergy bonus, plus a few others at level 30 (1%xp bonus for each). Legendary quests are really the way to go from 20-60, hopefully that changes because...

I would get the 3DS Circle pad pro for the second analog stick if you go the 3DS route. Helps give it that Wii U feel. If you're interested in MH4U then you should definitely just go the 3DS way to prepare yourself. That said, the game is pretty baller on the Wii U and looks great.
